I've gone longer without driving it in the past and when it was cold so this was a little odd. I assumed it would go away, but after driving 2 hours (1 hour each way) the light My ML320 is not shifting gears. transmission has gone into limp home mode. There is not much that you can do to make the transmission shift gears if it has gone into limp home mode. You cannot drive 300 miles in limp home mode. You are going to need to have a diagnostic scan performed to check the fault codes in the transmission control unit.
