My ML320 is not shifting gears. transmission has gone into limp home mode. There is not much that you can do to make the transmission shift gears if it has gone into limp home mode. You cannot drive 300 miles in limp home mode. You are going to need to have a diagnostic scan performed to check the fault codes in the transmission control unit.

The other leaks may be caused by a clogged oil vapor recovery system (PCV). Have them check and make sure that there isnt excessive air pressure building in the crankcase. 2008 ML320 CDI. Iridium Silver/Black MBTex, P2 pkg, NAV, Trailer Hitch. Purchased 3/2012 CPO 142,000 miles.
There's a number of conditions that need to be correct for the engine to start. One is the crankshaft position sensor which was a known problem on some 2008 CDIs. It was a recall/TSB item. If yours was subject to the recall and replaced then the next thing that comes to mind are the camshaft position sensors.

The 400 A fuse is from the pre-fusebox to the alternator, protecting a 35 mm² wire. The glow output stage is powered from a joint to this 35 mm² wire. This feed wire itself is 10 mm² and likely catches fire before the 400 A fuse blows up. Again one could blame the design but blaming the design does not make the problem go away.
